Harlan GenCheck™ QPCR

Quantitative PCR (QPCR) is employed for all zygosity testing within Harlan GenScreen™. The same stringency and internal testing is employed to validate DNA quality prior to quantitative measurements. QPCR is used to measure a fluorescent signal after every PCR cycle allowing for analysis of data in the logarithmic phase, not at the end point where the PCR products can no longer be quantified. For this application, an endogenous single copy gene is used for data normalization required to measure the relative copy number of the gene of interest. To accomplish the most sensitive and specific detection, we employ Taqman® chemistry utilizing minor groove binding (MGB) probes to ensure success for both easy and challenging target sequences. Genotyping determinations are made using validated analytical tools, among them comparative Ct analysis.
