Overview

If you require a peptide (Amino Acid Sequence) to be synthesized, you need to provide the desired sequence so we can produce this to a purity of 85% or 95%. In case the peptide has a long sequence e.g.: 269mer with certain sections of interest we can look into this and come up with the most suitable Antigenic Peptide Sequences and once we have decided which would be more suitable for your needs the synthesis can begin. It is our standard procedure to synthesize 10mg of Peptide, 5mg of this is conjugated to KLH and 5mg left over for any further testing you may require. This is shipped to you at the end of the project if not required beforehand. 

Synthesis

  • Peptide synthesis is performed using an insoluble resin support with a linker attached. This can then be cleaved by TFA () to ensure that the amino acids couple in the correct orientation the various reactive groups of inappropriate reactions are protected. These are simultaneously removed when the peptide is cleaved from the solid support by the trifluoroacetic acid.

 Purification

  • Additional chemicals, termed scavengers are dissolved in the TFA. These absorb the released reactive species which could affect the purity of the peptide. The peptide may also contain truncated and deletion sequences which may interfere with activity of the peptide. Both truncated and deletion sequences and scavengers can be removed using purification by reverse-phase hplc. For antibody production purities of greater than 80% are advised.

 Analysis

  • All peptides are analysed for purity by hplc and mass spectroscopy. The hplc trace is supplied with each peptide, and amino acid analysis can be provided on request.

 Presentation and Storage

  • Synthesised purified peptides are supplied as lyophilized powders. Peptides should be stored in a sealed glass vial protected from light at –20Cº with a desiccant. Avoid repeated freeze/thaws. Harlan Laboratories reserves the right to review all peptides before synthesis. If a peptide is determined to require special procedures, a separate quotation will be issued before synthesis begins. For conjugation of your peptide to KLH or other carrier molecules, we refer to the conjugation section.

  • If you require modification of your peptide such as cyclisation or phosphorylation please contact us for an individual quote. Harlan offers peptide synthesis using continuous flow Fmoc (9-fluorenylmethyloxycarbonyl) chemistry.
