Harlan Bioproducts for Science, Inc. offers a custom-collected serum for use in Whole Embryo Culture (WEC) of 7 to 9 day old rat and mouse embryos.

Whole Embryo Culture Serum has been used in culture assays as a predictive model for identifying teratogenic compounds that are often difficult to model with in-vitro systems.

It has been shown that early organogenesis stage rat embryos incubated with known teratogenic compounds in-vitro and cultured with Whole Embryo Culture Serum can provide both specific and sensitive morphological endpoints for identifying compounds that induce these defects in-vivo.

Fetal Bovine Serum

Product Description---Fetal Bovine Serum Premium Grade (Fetal Bovine Serum-Premium) is derived from the fetuses of healthy cattle. This high-quality product exhibits low hemoglobin, low endotoxin levels, and excellent growth-stimulating properties. Fetal Bovine Serum-Premium has not been physically altered, chemically treated, or irradiated. The final product is a normal, off-clot serum which has been sterilized through a series of three 0.1um filters.

Product Usage---Fetal Bovine Serum-Premium can be used for growth stimulation/promotion of a wide variety of primary tissue culture cells and established cell lines. The serum concentration needed for growth promotion varies from cell to cell. For most cells, the concentration ranges from 5% to 20%.

Physicochemical Analyses

Endotoxin-Product has an endotoxin level of less than 1ng/ml using the LAL gel clot method.
Hemoglobin-Product has a hemoglobin concentration of less than 20mg/dl.
Osmolality-Ranges between 260 and 320 mOsm/kg.
Total Protein-Protein concentration is determined by the Biuret method and ranges between 3.2 and 4.5gm/dl.
pH-Ranges between 6.8 and 7.8.
Biochemical Profile-20 chemical analyses
-a chemical panel- is performed on all lots.
Immunoglobulin Concentration-An electrophoretic profile shows the distribution of serum proteins. The immunoglobulin G concentration is quantitated by a specific radial immunodiffusion assay.

Product Performance---All lots of Fetal Bovine Serum are tested for growth promotion with a wide variety of cell lines. Relative growth is determined by comparison with an in-house Fetal Bovine Serum standard.

Comparisons are made for growth promotion, doubling time, and plating efficiency. Tests are carried out after three passages of the cells in the test serum and the Fetal Bovine Serum standard. Throughout the testing procedure, cells are observed microscopically for any cytopathogenic effects. In addition, each lot is tested for its ability to support terminal dilutions of hybridoma cell lines.

Cytotoxicity---Each lot of Fetal Bovine Serum-Premium is free of any detectable cytotoxic effect on the test cell lines.
